在其他经验贴查到很多杂志要求整张膜敷抗体的图像，那就意味着western blot转膜结束后要敷目的显影，而后… 它是我们WB实验过程中很重要的一个参考系。通过内参结果，我们可以判断：1）实验流程和操作是否正常；2）实验中各组样本上样量是否一致；3）目标蛋白的表达水平是否存在差异。因此内参还被称为上样参照（Loading Control）。 一、 内参选择原则 昨天刚进实验室的小师妹跑来找小捷。她正想做个新的WB实验，但她刚进实验室还没有自己设计过WB，就来和小捷确认一下protocol。果不其然，师妹不知道该怎么使用抗体，只是模模糊糊地知道抗体需要稀释。可问到要稀释多少倍、用什么稀释，她可就只剩挠头了。 我们在阅读文献时所看到的WB实验图片中，也偶尔会出现双条带或多条带。这些现象是研究过程中值得解释和讨论的。在进行Western Blot实验时，我们应该清楚，针对某些目标蛋白的检测结果并不一定是只有一条带。 wb结果中可能出现有目标蛋白条带，但是目标条带在膜上颜色较浅，如下图。该现象原因与上面的无目标条带较为相似，可能导致该现象的原因我们也从4个方面进行分析，并给出相应的解决方法，具体分析见下表。 相机上的“wb”就是白平衡的参数设置键，在相机上表示出来用“wb”键来表示。. 白平衡的设置是根据拍照的天气和光线情况来选择，相机有很多模式，如日光、阴影、阴天、钨丝灯、白色荧光灯、闪光灯、阴影等等，供我们在不同的场景中选择使用。 首先恭喜你有一个wb非常好的抗体，毕竟能获得做wb非常好的抗体也不容易。其次，你这个抗体很可能是通过人工合成多肽得到的，和上面所述，你用来做抗原的多肽恰好被包埋在蛋白质的中心，因此只能识别wb中线性的部分。 q3. 如何选择免疫组化抗体？ 我跑的这个蛋白分子量是289、220kda 分子量应该算是偏大了，普通的marker也就到250 我从wb蛋白电泳的步骤来讲这个蛋白咋跑吧 首先是制胶，选择8%的胶可以把这个蛋白跑出来，而且稳定，条带清晰。 将目标蛋白条带发出的信号使用软件进行分析，计算出目标蛋白的相对定量值，即为wb定量。 对wb条带进行定量方法之一是灰度分析，灰度是计算机图像分析仪根据图像颜色深浅分为256个级别。灰度值越小物体颜色越深。 1、光密度与灰度 qrt-pcr 与wb是可以互换的实验吗？ 荧光定量PCR做的是研究基因的RNA层面的定量，WB做的是蛋白的定量，但是RNA本身也是可以翻译成蛋白的，所以是不是说这两个实验理论上来说是可以互换的…

2024.11.30 12:39 Dockers-Man Secretlab or alternate riser?

Hey all, I'll be setting up 3 monitors on my Magnus Pro XL, using one single monitor arm and one dual monitor arm. To achieve the desired height of the monitors I'll be attaching these directly to the desktop (as opposed to the riser).
Reviews suggest that the Secretlab riser and monitor arms may conflict with each other where they attach at the cable tray.
Does anyone have suggestions around how to use a desktop riser when using two monitor arms, as I don't plan to attach these to the riser?
A different brand riser could be an option?

I’m socially anxious, introverted, and shy. To overcome this, I force myself to talk to people at work or even to strangers. Even if I decide "I'll speak properly this time" I get the same result. My voice becomes quiet and very soft spoken, I stutter and mumble. I often go blank, especially when someone greets me. Sometimes I somehow manage to speak properly, but I don’t know how I do it
I get easily intimidated, even when speaking to someone shorter or physically weaker than me, as long as they are more confident or assertive. In rare cases, if I get shouted at, I almost feel like I might pee myself, I'm not even kidding
Therapy is not an option for me right now.
What can I do to improve this?

The inibuilds A320neo was a dream in 2020. Flew perfectly and the buttons works. The controls were intuitive even on xbox.
It seems Microsoft have ignored the old adage of ‘if it’s not broken don’t fix it’! The new controls mapping is just weird. Absolutely helpful quick keys no longer exist but my biggest complaint is the AP. NAV mode isn’t working correctly but can be overcome. The worst element is Altitude controls. It just will not engage managed mode. I’m not touch v/s at all so expect a steady climb in line with the MCDU but it’s deciding to climb like hell until stall speed is hit then drop like a stone. Why, why why??!!
Anyone got any suggestions for what I may be doing wrong given I had none of these issues in 2020?
