昨天刚进实验室的小师妹跑来找小捷。她正想做个新的WB实验，但她刚进实验室还没有自己设计过WB，就来和小捷确认一下protocol。果不其然，师妹不知道该怎么使用抗体，只是模模糊糊地知道抗体需要稀释。可问到要稀释多少倍、用什么稀释，她可就只剩挠头了。 我跑的这个蛋白分子量是289、220kda 分子量应该算是偏大了，普通的marker也就到250 我从wb蛋白电泳的步骤来讲这个蛋白咋跑吧 首先是制胶，选择8%的胶可以把这个蛋白跑出来，而且稳定，条带清晰。 qrt-pcr 与wb是可以互换的实验吗？ 荧光定量PCR做的是研究基因的RNA层面的定量，WB做的是蛋白的定量，但是RNA本身也是可以翻译成蛋白的，所以是不是说这两个实验理论上来说是可以互换的… 带着这些疑问，小编带大家梳理一下wb的蛋白定量。 为什么要进行蛋白定量？ goodWB实验需要对样品总蛋白进行定量，主要目的是对样品进行均一化处理，也就是使各个泳道蛋白总上样量一致。 首先恭喜你有一个wb非常好的抗体，毕竟能获得做wb非常好的抗体也不容易。其次，你这个抗体很可能是通过人工合成多肽得到的，和上面所述，你用来做抗原的多肽恰好被包埋在蛋白质的中心，因此只能识别wb中线性的部分。 q3. 如何选择免疫组化抗体？ 将目标蛋白条带发出的信号使用软件进行分析，计算出目标蛋白的相对定量值，即为wb定量。 对wb条带进行定量方法之一是灰度分析，灰度是计算机图像分析仪根据图像颜色深浅分为256个级别。灰度值越小物体颜色越深。 1、光密度与灰度 首先，做好个人防护，口罩，橡胶或者丁腈手套，确认没有破裂，穿工作服，不穿露脚的鞋子。 它是我们WB实验过程中很重要的一个参考系。通过内参结果，我们可以判断：1）实验流程和操作是否正常；2）实验中各组样本上样量是否一致；3）目标蛋白的表达水平是否存在差异。因此内参还被称为上样参照（Loading Control）。 一、 内参选择原则 wb结果中可能出现有目标蛋白条带，但是目标条带在膜上颜色较浅，如下图。该现象原因与上面的无目标条带较为相似，可能导致该现象的原因我们也从4个方面进行分析，并给出相应的解决方法，具体分析见下表。 wb显影条带太淡的情况，通常考虑两个方便：①显影过程中信号放大的问题；②蛋白样品上样量不够。 ①显影过程中信号放大的问题 这部分又可以划分为多类情况，主要为：一抗-二抗信号级联（信号差，条带弱，有可能会是一抗或者二抗的使用浓度不够，大 ...

Pretty much as the title says, I’m a Irish guy living in England at the moment but goes between here and Ireland a lot, just looking for some connections. Now we’ve that out of the ways let me tell you about myself -

You - I tend to be attracted people who are “Thic” alt women who have tattoos and piercings but definitely not a dealbreaker. I have a Nighthawk MK83 mesh system with a AX3600 router and 3 satellites in my home. I've noticed that some of my devices on one side of my home aren't working too well, so I opened the Nighthawk app and poked around a bit. Turns out that every device in my home is connected to the router, and none of them are connected to any of the satellites. Even though the satellites are significantly closer to the devices. And yes, all of the satellites are paired, have blue lights, and look like they're working in the app. I've tried cycling power on everything but no luck. Any ideas?
