Hii! I am not sure if this is the right place to post this but i am doing my masters thesis and I am working with the cell line MIA PaCa-2. I've been taught how to maintain a cell line. How to passage, cell count (haemocytometer). Recently I started the work with MTT (24hrs) to figure out the IC50 of my drugs. The first time I did it, a few PhD Scholars told me my cells didn't die. Cell viability for some of the drug concentration was better than control. But this work has been done already and the PhD Scholars knows the result so they asked me to repeat it this time with 24hrs and 48hrs. It also failed. My cells aren't dying. I feel stupid that I can't even do something so basic. One thing I keep noticing is that the day I add MTT reagent, when I am aspirating the 200ul (complete media+drug), the volume is less. Around 20 or 30ul from every single well. I add 100ul media during seeding and then on the 2nd day during drug treatment I add 100ul complete media for control, 80ul complete media for drug (single concentration) and 60ul complete media (drug combination). Then 20ul of drug. I am so sure I am adding 100ul on the second day but still! Is that normal? Is it evaporation? Is it my pipetting error? After the 4hrs incubation of mtt, when I am aspirating before to add DMSO, the volume is correct (100ul media and 20ul mtt reagent). I am scared to ask others because shouldn't I already know all this. One of the Scholars told me it might be due to cell counting mistake or drug not properly mixed. They checked my counting and told me it's not the problem. I vortex each of the drug concentration during serial dilution. I mix it with my pipette also. I mix it with pipette before adding to the 96 wells. They told me it's okay, you're doing it for the first time. I'm doing MTT again today at 4. I know it's gonna failðŸ˜
What might be going wrong? What can I do to fix this?

I bought some new clippers to hopefully save on cutting my hair. I got some Andis Beauty Master+ and was so excited when they showed up in the mail. I was disappointed to see that the oil despite being placed in a small bag leaked everywhere in the kit. It did manage to get onto the clippers despite them being wrapped in a bag as well. I'm worried that the oil has gotten into the buttons potentially damaging the clippers. Before testing them out should I just ask for a exchange of new clippers? Also any advice on cleaning them? I already contacted support and they offered a new bottle of oil but nothing else. I think if I ask for a return they will oblige.
